چکیده

OBJECTIVE To investigate the effect of neurogulin-1 (NRG1) on the transmission and plasticity of CA1 synapses in mouse hippocampal slices at different temperatures. METHODS Under room temperature (26-/+1 degrees C) or physiological temperature (32-/+1 degrees C), field excitatory postsynaptic potential (fEPSP) evoked by extracellular microelectrode recording technique was recorded in the CA1 region in adult mouse hippocampal slices, and the long-term potentiation (LTP) was induced by high frequency stimulation (HFS). The effects of NRG1 on the baseline fEPSP and induction of LTP in CA1 region were observed. RESULTS No significant variation in the slope of fEPSP relative to the baseline fEPSP was observed after perfusion with NRG1 at room temperature or physiological temperature (P > 0.05). After HFS at the room temperature, the mean slope of fEPSP in the slices perfused with NRG1 was similar to that of the control group (P > 0.05), but HFS at the physiological temperature resulted in significant reduction in the mean slope of fEPSP in the slices perfused with NRG1 (P < 0.01). CONCLUSION NRG1 may temperature-dependently inhibit the induction of LTP in the CA1 region in mouse hippocampal slices.

Role of adenosine receptors and protein phosphatases in the reversal of pentylenetetrazol-induced potentiation phenomenon by theta pulse stimulation in the CA1 region of rat hippocampal slices

The effect of theta pulse stimulation (TPS) on pentylenetetrazol (PTZ)-induced long-term potentiation of population spikes (PS) was studied in the hippocampal CA1 in vitro. A transient PTZ application produced a long-lasting enhancement of PS amplitude. A 3-min episode of TPS delivered at a higher intensity produced complete reversal of the PTZ potentiation when delivered during the last minute...

Copper sulfate inhibits seizure activity induced by pentylenetetrazole in mice

Background and Objective: Copper is one of the main micronutrients of body which plays a key role as a cofactor in the function of metabolic enzymes. Previous studies have shown that copper sulfate () inhibits long-term potentiation (LTP) in slices of hippocampal CA1 region. Whereas LTP is involved in learning and epilepsy, it seems that copper effects on LTP could be associated with its effect...

Cysteamine pretreatment reduces Mg2+-free medium-induced plasticity in the CA1 region of the rat hippocampal slices

Extracellular population responses are largely preferred for the study of long-term potentiation (LTP). The effect of Mg2+-free medium on changes in activity and plasticity of Schaffer collateral-CA1 pyramidal cell synapses was examined. Hippcampal slices from cysteamine-treated (200 mg/kg, s.c.) and saline-injected (1 ml/kg, s.c.) albino rats were perfused with ACSF. Population spikes (PS) wer...

Role of NMDA receptors and voltage-dependent calcium channels in augmenting long-term potentiation of the CA1 area in morphine-dependent rats

The involvement of NMDA receptors and voltage-dependent calcium channels in augmentation of long-term potentiation (LTP) was investigated at the Schaffer collateral CA1 pyramidal cell synapses in hippocampal slices of morphine dependent rats, using primed-burst tetanic simulation. The amplitude of the population spike and its delay were measured as indices of increase in postsynaptic excitabi...
